I constructed a safe in my bedroom for my hearthfire home. Checking the container in the console, it shows the container is set to respawn. No limit to joining guild. The only limitation is Stormcloaks/Imperials. If you are in the Dark Brotherhood, you can join still join the Thieve's Guild, etc. Also, a quick word, the Dark Brotherhood quests are by far my favorite. Have fun!
